摘要
目的建立测定多索茶碱血浓度的反相高效液相色谱法。方法色谱柱为UltrasphereC18柱(250mm×4.6mm,5μm),流动相为乙腈-水(梯度洗脱),流速为1.0mL/min,柱温为室温,检测波长为210nm。结果多索茶碱质量浓度在0.39~25.00mg/L范围内,与多索茶碱跟内标峰面积比值的线性关系良好,回归方程为Y=7.5098X-0.1300,r=0.9998;多索茶碱的最低检测质量浓度为0.2mg/L(S/N≥3);平均回收率为104.4%,日内、日间精密度RSD均小于8%。结论该法可用于多索茶碱血浓度的测定。
Objective To establish a method for determining the concentration of doxofylline in serum by RP-HPLC.Methods The column was Ultrasphere C18(250 mm × 4.6 mm,5 μm),the mobile phase was acetonitrile-water,the detection wavelength was 210 nm,the flow rate was 1.0 mL /min,the temperature of column was at room temperature.Results The calibration curves were linear in the range of 0.39-25 mg /L for doxofylline,the linear regression equation was Y = 7.509 8 X-0.130 0(r = 0.999 8),and the limit of determination was 0.2 mg /L,the average recovery rate was 104.4%,and RSD of intra-day and inter-day was below 8%.Conclusion This method is usable for monitoring of doxofylline.
