预处理和后处理对大鼠后肢缺血—再灌注肾损伤的保护作用

The protective effect of ischemic pre-conditioning and post-conditioning on the kidney injury in hind limb ischemia reperfusion of rat model

摘要 目的观察肢体缺血预处理、后处理对大鼠后肢缺血—再灌注肾损伤的保护作用。方法取雄性Wistar大鼠24只,建立后肢肌肉缺血—再灌注模型,实验分为4组,每组6只,A组为假手术组,只进行常规解剖不阻断血管,15 h处理;B组为缺血—再灌注组,缺血6 h,再灌注9 h;C组为缺血前给予10 min缺血,10 min灌注,重复3次后行缺血5.5 h,再灌注8.5 h组;D组为缺血5 h 57 min,再灌注前给予1 min灌注,1 min缺血,重复3次后继续再灌注8 h 57 min。各组大鼠均取血清检测乳酸脱氢酶(LDH)、肌酐(Cr)含量及肾组织匀浆丙二醛(MDA)含量,取肾组织石蜡包埋,HE染色,观察其病理形态学变化。结果 B、C、D组上述指标均明显高于A组(P<0.01),C、D组明显低于B组(P<0.01),C组与D组比较差异无统计学意义(P>0.05)。A组组织形态基本正常,C、D组组织形态学改变较B组明显减轻。结论肢体缺血预处理、后处理均可减轻实验大鼠后肢缺血—再灌注肾损伤,且无差异。 Objective To observe the protective effect of ischemic pre-conditioning and post-conditioning on the kidney injury in hind limb i.schemia reperfusion(I/R) of rats model.Methods Twenty four male Wister rats were enrolled to establish I/R model.Thev were divided into four groups.A is sham operation group;B is I/R group;C is I/R group with ten minutes ischemia,ten minutes reperfusion and repeating three times before 5.5 h ischemia; D is I/R group with one minute ischemia,one minute reperfusion and repeating three limes before 8 h 57min reperfusion.Test of serum LDH,Cr and the level of MDA in kidney tissue after reperfusion were performed,meanwhile, the morphologic changes were obverse.Results Serum level of LDH,Cr and the contents of MDA in group B,C,D were higher than in group A(P0.01 ),it was lower in group C and group D than that in group B(P0.01).There was no difference between group C and group D(P0.05).Morphological change of group B were server than that in group C and group D,and there is no obvious morphological changes in group A.Conclusion Ischemic pre-conditioning and post-conditioning may inhibit the kidney injury in hind limb ischemia-reperfusion(I/R) of rats model.There is no difference of effect between ischemic pre-conditioning and ischemic post-conditioning.
