摘要
目的研究抗菌蛋白血小板型磷脂酶A2(PLA2)、杀菌/渗透增强蛋白(BPI)和β-防御素-1mRNA在正常大鼠生殖系统内的存在情况,以了解生殖系统的天然抗菌机制的状况。方法从大鼠生殖系统不同器官和组织的匀浆中提取总RNA,合成第1链cDNA,利用PCR扩增大鼠血小板型PLA2、BPI和β-防御素-1的DNA片段,在凝胶电泳的胶上观察PCR产物条带。结果在大鼠的7种生殖器官和组织中,睾丸、附睾和卵巢均能检测到血小板型PLA2、BPI和β-防御素-1的mRNA,其中以卵巢中的血小板型PLA2mRNA、睾丸中的BPImRNA和附睾中的β-防御素-1mRNA含量尤为丰富;另外,在前列腺、子宫、阴道中也能检测到β-防御素-1和血小板型PLA2的mRNA。结论血小板型PLA2、BPI和β-防御素-1在大鼠雄性和雌性生殖系统中的多种器官和组织存在,提示天然免疫的重要成分抗菌蛋白在防御生殖系感染方面起了重要的作用。
OBJECTIVE To investigate the existence of bactericidal/permeability increasing protein (BPI), β-defensins-1 and platelet phospholipase A2 (PLA2) mRNA in normal rat genital system, for providing important information about anti-bactericidal protein in clinical application. METHODS The total RNA was extracted from organs and tissues homogenates. Then the first-strand cDNA was synthesized and BPI or β-defensins 1 or PLA2 DNA was amplified by PCR. RESULTS BPI,β-defensins-1 and PLA2 mRNA were found in testis, epididymis and ovary from 7 various genital organs and tissues. Among them, BPI mRNA of testis, β-defensins-1 mRNA of epididymis and PLA2 mRNA of ovsary were rather aboundant. Moreover,β-defensins-1 and PLA2 mRNA were also found in prostatic gland, uterus and vsagina. CONCLUSIONS BPI,β-defensins-1 and PLA2 mRNA are present in many organs and tissues of rat genital system, which indicate that anti-bactericidal proteins take important part in host defense against bacterial infection.
