摘要
为探索柔毛霉素(daunorubicin,DNR)、骆驼蓬总碱(totalalkaloidofHarmaline,TAH)对兔后发性白内障形成的影响及对兔眼的毒性作用。于兔晶体囊外摘除(extracapsularlensextraction)术中分别注入0.1ml的TAH(0.2mg/ml)、DNR(0.2mg/ml),观察兔晶体后囊混浊、眼内压的变化等;并观察术后组织病理学和超微结构的变化。结果显示:15眼注入TAH术后炎性反应与对照眼无明显差别;术后2周后囊膜明显比对照组清晰,术后4周有轻度混浊,但比对照眼轻。至术后8周仍有差别;10眼注入DNR后,表现出明显的炎性反应,角膜明显水肿且增厚,前房大量纤维素性渗出。组织病理学检查表明,对照眼术后晶体上皮细胞(lensepithelialcels,LEC)增生,并形成明显的soemmeting环。TAH引起晶体上皮细胞空泡变性、核固缩并能明显抑制兔眼的晶体上皮细胞增殖,且对角膜内皮及其它组织无明显毒性;而DNR虽可明显抑制LEC增生,但角膜基质层明显水肿并伴有大量的炎性细胞浸润,且角膜内皮细胞均很少见,表现了明显的眼部毒性。结论:TAH能有效的抑制晶体上皮细?
piroose:To investigate the effect of daunorubicin(DNR)and total alkaloid of Harmaline(TAH)as therapeutic agent whether it is beneficial in prevention of posterior capsular opacificatoion(PCO)and the toxicity of these drugs.Methods:Fifteen rabbit eyes were injected 0.1ml of 0 2mg/mlTAHand ten eyes 0 2mg/mlDNR into the capsular bag during extracapsular lens extraction(ECLE)respectively.The intraocular pressure(IOP)were measured,the postrerior capsule were observed by biomicroscopy.The histopathological changes and transmmision electronic microscope examination were also studied.Results:The inflammatory response was mild in both TAH and control groups after injection,but were more severe in DNR and group ln DNR group all eyes showed severe corneal edema and opacity.The corneal edema lasted for 4 to 8 weeks.PCO formation occured both in TAHtreated and control group by 4 weeks,however,it was much milder in the TAHtreated eyes;because of corneal opacity it could not observed the PCO by biomicroscope in DNR group eyes.IOP at first day after operation was higher than before and 2nd and 3rd day aftet operation.Histopathological study revealed that lens epithelial cells proliferated and Soemmering`s ring developed in the control eyes,however,only mild vacuolization and pkynotic changes of lens epithelial cells was noted in the TAH treated eyes.In DNR group there were obvious cornel edema and esophobic infiltration;the corneal endothelial cells were severely lost;however,in the capsular bag,there were no lens epithelial cells proliferation and lens cortex remain.Transmmission electronic microscopy demonstrated that there were mild cytoplasmic vacuolization,mitochondrial swelling in lens epithelial cells in TAH group.Conclusion:The results suggest that Alt could inhibit metaplasia and proliferation of lens epithelial cells in rabbit eye without obvious toxicity,it may be used as a potential agent in the prevent the development of PCO.DNR could obviously inhibited lens epithelial cells proliferation,but because of severe toxicity it must be further studied about the method of administration and the concentration.
