Staurosporine对人低分化鼻咽癌细胞系细胞周期和DNA倍体含量影响的动态观察

摘要 目的:探讨蛋白激酶C(PKC)抑制剂Staurosporine(ST)诱导CNE-2Z细胞凋亡时,对细胞周期的选择性和对DNA含量及细胞周期的动态影响。方法:以PKC催化区抑制剂ST作用于CNE-2Z细胞不同时间后,收集细胞进行PI染色和流式细胞仪检测。结果:ST以2×10-6mol/L终浓度分别作用至3,6,12,24h后,发现G2期百分比分别为30.43±7.16、30.83±6.25、42.43±3.98和71.80±7.55,均较对照组12.08±3.49增高明显。比较差异有显著性(P<0.01);而G1期在3,6,12,24h分别为34.70±1.56,37.03±6.47,16.63±1.77和15.30±5.46,均较对照组62.56±6.57明显降低,比较差异有显著性(P<0.01)。亚二倍体峰百分比在12、24h分别为37.10±7.21和43.72±6.73,均较对照组9.81±1.78显著增加(P<0.01);而12h以前该峰增加不明显。结论:ST在终浓度为2×10-6mol/L诱导CNE-2Z细胞至12h后即可出现明显的DNA含量降低的凋亡细胞,对G2期细胞敏感而使之逐渐阻滞,且存在? Objective:To investigate the changes of cell cycle and DNA diploid in CNE 2Z cells induced by the inhibitor of protein kinase C(PKC).Methods:CNE 2Z cells were treated by PKC inhibitor staurosporine(ST),acting on the catalytic domain,at the concentration of 2×10 -6 mol/L for various hours,stained with propidium iodide (PI),and tested by flowcytometry (FCM).Results:Within 3,6,12,and 24 hours,the percentages of G 2 phase were 30.43±7.16,30.83±6.25, 42.43 ±3.98 and 71.80±7.55 respectively, all of which were higher than that of control groups 12.08±3.49( P < 0.01). The percentages of G 1 phage were 34.70±1.56,37.03±7.21,16.63±1.77,and 15.30±5.46 respectively, all of which were lower than that of control groups 62.56±6.57( P <0.01).The percentage of hypodiploid was 37.10±7.21 and 43.72±6.73 for 12 and 24 hours respectively,and both obviously increased compared with that of control groups 9.81 ±1.78( P <0.01).Conclusion:The results showed that the number of apoptotic cell induced by ST at the concentration of 2×10 -6 mol/L gradually increased while the G 2 phase increasing and G 1 phase decreasing within 24 hours,and it suggested that ST could induce CNE 2Z cells apoptosis and selectively affect G 2 phase of cell cycle.
