摘要
用浸泡法和明胶包裹法制备庆大霉素重组合异种骨,植入小鼠股部肌袋中,用高效液相色谱法测定不同时间移植骨粒周围软组织中庆大霉素浓度,观察庆大霉素重组合异种骨复合物的药物释放度。结果表明:①抗生素快速释放时相持续12~24h,而后释放量缓慢逐步递减;②明胶包裹4mg的庆大霉素重组合异种骨复合物具有良好的药物缓释功能,在植入部位周围软组织中维持有效浓度达17天;③提高明胶浓度能进一步延缓药物释放度。
Open fractures are often contaminated and the development of infection is a major complication. Infection is favored by revitalization of bone and soft tissue and loss of skeletal stability. Maintenance of therapeutic tissue levels of antibiotic to target areas requires high serum level of potentially toxic and costly antibiotics. In order to solve the problems of bone defects and infection all at once, we prepared the gentamicin loaded reconstituted bone xenograft (G RBX) aiming at putting local administration of antibiotics and bone transplantation into one step. The results showed that the initial rapid release of antibiotic lasted 12 ̄24 hours, and it was followed by a second phase of sustained but gradually diminishing dissolution. Effective release of gentamicin from G RBX without gelatin maintained only for 5 days in mice, but for 17 days in G RBX encapsulated with 40% gelatin. The rate of release can be slowed down by increasing the concentration of gelatin.
