摘要 目的研究难治性癫痫细胞模型中lamininβ1表达的变化,探讨lamininβ1在难治性癫痫形成中的作用。方法 SD新生乳鼠,迅速断头后取出双侧海马,体外培养海马神经元至第9天,随机分为对照组和模型组,采用荧光定量PCR检测两组12h、24h和48h lamininβ1mRNA表达,免疫组化技术检测lamininβ1蛋白的表达。结果荧光定量PCR显示在12h后模型组lamininβ1mRNA表达增加(P<0.05),且随着时间延长表达增强。在24h,免疫组化显示模型组lamininβ1蛋白表达增强(平均光密度值:对照组0.0947±0.0259vs模型组0.1351±0.0262,P<0.01).结论难治性癫痫细胞模型中lamininβ1表达增加,提示lamininβ1可能参与难治性癫痫的形成过程。 Objective To study the change of lamininβ1 gene expression in cell model of intractable epilepsy,and to explore the role of lamininβ1 in this model's formation.Methods The neonate SD rats were decapitated quickly to obtain the hippocampal neurons,which were cultured in vitro until 9 days and randomly divided into control group and model group.Expression of lamininβ1 mRNA were examined by FQ-PCR at the time of 12h,24h and 48h after the model established,and the expression intensity of lamininβ1 protein in two groups was examined by immunohistochemistry.Results Through FQ-PCR,after 12h,the expression of lamininβ1 mRNA in model group increased over time (P0.05).At 24h,the immunohistochemistry displayed the expression intensity of lamininβ1 protein increased in the model group(AOD:control group 0.0947±0.0259 vs model group 0.1351±0.0262,P0.01).Conclusion The expression of lamininβ1 was upgraded in intractable epilepsy cell model,which might be an important factor for the model's formation.
