摘要
目的探讨肝癌腹水细胞膜表面糖链表达谱。方法用H22细胞制备肝腹水模型鼠,36只小鼠随机分为正常组(6只)和肝腹水组(20只),注射10d后抽取腹水提取癌细胞用吖啶橙荧光标记,应用凝集素芯片检测细胞膜表面糖表达谱。结果肝癌腹水细胞膜表面有大量唾液酸、乙酰葡萄糖、乙酰半乳糖、甘露糖和半乳糖糖链表达,正常小鼠肝组织中只有多聚乳糖胺类型糖链表达。结论肝癌发生过程中伴随着细胞膜糖链类型和数量增加,唾液酸、乙酰葡萄糖、乙酰半乳糖、甘露糖和半乳糖糖链表达增加与肝癌变发生发展紧密相关。
【Objective】To investigate the changes and important roles of glycoprofiling on cell surface from ascites of liver cancer in mice.【Method】 Ascites mice were induced by injecting H22 cells into mice abdomen.Thirty-six mice were randomly divided into normal group(6 mice) and ascites group(20 mice).After 10 days the ascetic fluid was collected to separate the tumor cells and liver cells were extracted from normal mice after digesting.All cells were labeled with Acridine Orange(AO) cell tracking dye and detected by lectin microarray to analyze the cell surface glycoprofiling.【Result】Sia,GluNAc,GalNAc,Man and Gal increased on cell surface from ascites,only polyLacNAc and Lac/LacNAc were expressed on normal liver cell surface.【Conclusion】 The glycosylation of cell surface is increased in occurrence of hepatocarcinogenesis.The increasing expression of Sia,GluNAc,GalNAc,Man and Gal may play key roles in liver carcinogenesis.
