摘要
目的探讨Transgelin基因在卵巢浆液性肿瘤发生、发展过程中的作用。方法采用逆转录-聚合酶链反应(RT-PCR)及WesternBlot方法对10例正常卵巢、11例卵巢良性浆液性瘤、14例卵巢交界性浆液性瘤及38例卵巢浆液性癌组织中Transgelin基因mRNA及蛋白进行检测。结果 Transgelin基因mRNA及蛋白在卵巢浆液性癌中阳性表达率及相对含量皆低于卵巢交界性浆液性瘤、良性浆液性瘤及正常卵巢组织,差异均有显著性(P<0.05)。Transgelin基因mRNA及蛋白低表达或缺失与卵巢浆液性癌的临床手术病理分期、组织学分级及淋巴结转移有关,与有无腹水无关。结论 Transgelin基因低表达或表达缺失为卵巢浆液性肿瘤发生过程中的早期事件,可能是卵巢浆液性肿瘤发生过程中的抑制因素,与卵巢浆液性癌的发生、发展密切相关。
【Objective】To investigate the expression of transgelin in ovarian serous carcinoma tissues and to understand its role in the tumorigenesis and progression of ovarian serous carcinoma.【Methods】 Reverse transcription polymerase chain reaction(RT-PCR) and Western blot were used to assess the expression of transgelin mRNA in 38 cases of ovarian serous carcinoma,14 cases of ovarian borderline serous tumors,11 cases of ovarian benign serous tumors and 10 cases of normal ovary tissues.【Results】Positive expression and relative content of transgelin mRNA and protein in ovarian serous carcinoma were statistically lower than those in ovarian borderline serous tumors,ovarian benign serous tumors and normal ovary tissues(P〈0.05).The expression of transgelin mRNA and protein were correlated with the pathological stage,grade and lymph node metastasis of cancer but not ascites.【Conclusions】Transgelin expression is low or lost in ovarian serous carcinoma.Transgelin maybe play a suppression role in tumorigenesis and progression of ovarian serous carcinoma.
