摘要
目的从基因水平探讨过氧化物酶增殖体激活受体γ2基因(PPARγ2)Pro12Ala多态性对2型糖尿病(T2DM)患者脂代谢的影响。方法选取辽宁地区2型糖尿病患者331例,采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术对PPARγ2基因Pro12Ala多态性的基因型进行基因检测,并测定血脂代谢的指标。结果 2型糖尿病患者中总胆固醇(TC)和低密度脂蛋白胆固醇(LDL-C)的水平Pro12Ala/Ala12Ala组>Pro12Pro组,差异具有显著性(P值分别为0.032和0.026)。将T2DM患者按肥胖组(BMI>25kg/m2,171例)和非肥胖组(BMI≤25kg/m2,160例)分层后TC、TG、LDL-C的水平Pro12Ala/Ala12Ala组>Pro12Pro组,差异具有显著性(P值分别为0.028、0.025和0.012)。按性别分层后未发现差异有显著性。结论 PPARγ2基因Pro12Ala多态性影响2型糖尿病血脂代谢(TC、LDL-C水平升高而对TG、HDL-C影响差异无显著性),在肥胖个体中影响更为显著(TC、TG和LDL-C水平在两组间差异均有显著性),而与性别无关。
Objective To investigate the influence of peroxisome proliferation activated receptorγ2 gene Pro12Ala Polymorphism to the serum lipid of type 2 diabetes mellitus (T2DM) in Chinese people. Methods 331 T2DM patients were selected. The polymorphism of PPARγPro12Ala was detected by PCR-RFLP. Compare the levels of serum lipids between Pro12Ala/ Ala12Ala group and Pro12 Pro group, then we divided them into two layers (BMI25 kg/m2, n =171 cases) and non-obese group (BM I≤25 kg/m2, n =160 cases), According to body-mass index (BMI). And then divided 331 T2DM patients into two layers according to sex to compare the levels of serum lipids between Pro12Ala/Ala12Ala group and Pro12 Pro group in each layer. ResultsThe serum lipid levels of TC and LDL-C were higher in Pro12Ala/Ala12Ala group than in Pro12 Pro group (P value was 0.032 and 0.026, respectively) in T2DM. especially in obese patients Pro12Ala/ Ala12Ala group, The levels of TC, TG and LDL-C were higher than those in Pro12 Pro group P value was 0.028, 0.025 and 0.012, respectively); Association is not found between PPARγ2 Gene Pro12Ala and sex. ConclusionPPAR-γ2 P12A gene polymorphism may affect the levels of serum lipids in T2DM, especially in obese patients, but association is not found with sex.
