IgA_1对肾足细胞分泌VEGF、Col-Ⅳ的影响及复方积雪草2号的干预作用 被引量:4

摘要 目的:探讨复方积雪草2号对IgA1诱导的小鼠肾足细胞分泌血管内皮生长因子(VEGF)、IV型胶原(Col-IV)的影响。方法:用体外实验的方法,将小鼠肾足细胞经培养及传代,分成正常组、模型组、复方积雪草2号高、中、低剂量组、泼尼松龙组及缬沙坦7组,用IgA1诱导并分别予以上述各药进行干预24小时,用RT-PCR及ELISA法分别检测VEGF、Col-IV的mRNA及其蛋白质的表达。结果:IgA1刺激可以使小鼠足细胞VEGF与Col-IV表达明显升高(P<0.05),泼尼松龙、缬沙坦和复方积雪草2号的干预均能明显降低小鼠足细胞VEGF的高表达,与模型组相比,P均<0.05;而且泼尼松龙和复方积雪草2号还能明显降低足细胞Col-IV的高表达,与模型组相比,P均<0.05。结论:复方积雪草2号可抑制肾足细胞由IgA1诱导造成的VEGF、Col-IV的高表达。 Objective:To explore the effects of compound jixuecao(herba centellae)2 on vascular endothelial growth factor(VEGF)and IV type collagen(Col-V)secreted by mice'podocytes induced by IgA1.Methods:Experiment in vitro,mice' podocytes were cultured and passaged,they were divided into normal,model,compound jixuecao 2 high,middle and low dose groups,prednisolone and valsantan groups.Podocytes were induced by IgA1 and intervented by corresponding medicine for 24 hours,VEGFmRNA and Col-IV mRNA and their protein expressions were determined by RT-PCR and ELISA methods.Results:The VEGFmRNA and Col-IV mRNA and their protein expressions were elevated obviously after IgA1 induced,which were lowered significantly after prednisolone,valsantan and compound jixuecao 2 respectively intervened;prednisolone and compound jixuecao 2 could significantly reduce the high expression of Col-IV expression,there was obvious difference between them and model group(P<0.05).Conclusion:Compound jixuecao 2 can inhibit the high expression of VEGF and Col-IV in mice' podocytes induced by IgA1.
