人参皂甙Rg1对TGF-β1诱导的肾小管上皮细胞增殖及细胞外基质的影响

摘要 目的:观察人参皂甙Rg1对转化生长因子-β1诱导的肾小管上皮细胞增殖及细胞外基质的影响。方法:MTT法测细胞增殖情况。ELISA法测定培养细胞上清液中Col-Ⅰ,Col-Ⅲ和FN的含量。结果:在10~40ng/mL浓度范围内时间、剂量依赖性的促进肾小管上皮细胞增殖,减少转化生长因子-β1诱导的肾小管上皮细胞对Col-Ⅰ、Col-Ⅲ、FN的分泌。结论:G-Rg1能够促进TGF-β1诱导下的肾小管上皮细胞的增殖,可能与抑制ECM的分泌有关。 Objective:To investigate the effect of panaxoside Rg1 on proliferation and extracellular matrix(ECM) production of rat renal tubular epithelial cells induced by TGF-β1 in vitro.Methods:Rat renal tubular epithelial cells were incubated with different concentrations of panaxoside Rg1 and the same concentration of TGF-β1.Cell proliferation was assessed by MTT colorimetric assay.Fiberonectin(FN) and type Ⅰcollagen and type Ⅲ collagen contents were detected by ELISA.Results:Administration of 10-40mmol/L panaxoside Rg1 into culture medium could significantly promote cell proliferation in time-dependent and dose-dependent manner.Panaxoside Rg1 also decreased the production of FN and Co1Ⅰand Ⅲ.Conclusion:Panaxoside Rg1 could significantly promote renal tubular epithelial cells proliferation,which could be related with suppressing ECM production induced by TGF-β1.
