南极细菌胞外多糖体外抗单纯疱疹病毒Ⅰ型的实验研究 被引量:1

Experimental study on anti-HSV-Ⅰactivity of the extracellular polysaccharides from Antarctic bacterium

摘要 目的探讨一种南极细菌胞外多糖体外抗单纯疱疹病毒Ⅰ型(HSV-Ⅰ)活性。方法采用CPE观察与MTT比色法测定南极细菌胞外多糖对人角膜上皮细胞(HCEC)的毒性作用和抗HSV-Ⅰ作用,计算细胞存活率、病毒抑制率及病毒滴度。结果南极细菌胞外多糖对HCEC细胞毒性小,在低浓度下明显促进细胞生长,12.5μg·mL^(-1)浓度下存活细胞量是细胞对照组的146.88%;对HSV-Ⅰ有明显的抑制作用,实验最佳病毒抑制浓度为200μg·mL^(-1)。结论此南极细菌胞外多糖具有显著的低毒、促进细胞生长和抗HSV-Ⅰ活性。 Objective To examine the anti-HSV-Ⅰ activity of the extracellular polysaccharides from Ant- arctic bacterium. Methods The toxicity of the extracellular polysaccharides on HCEC and its anti-herpes simplex virus activity were assessed by CPE assay and MTT assay. The HCEC survival rates, HSV-Ⅰ inhibition rates of the extracellular polysaccharides and virus titer were calculated. Results The extracellular polysaccharides had little cytotoxic effect on HCEC, and it could promote cell's growth significantly in low concentration, the cell survival rate was 146.88% at dose of 12. 5 μg·mL^-1 extracellular polysaccharides. The extracellular polysaccharides could significantly inhibit HSV-Ⅰ replication, and the optimum inhibition concentration was 200μg·mL^-1. Conclusion The extracellular polysaccharides from Antarctic bacterium exhibits apparent low toxicity and anti-HSV-Ⅰ activity, and promotes cell's growth.
