沙立度胺及其对映体对人肺癌细胞系NCI-H460细胞抑制作用 被引量:1

Inhibition of Thalidomide and Its Enantiomer on Human Lung Carcinoma Cell Line NCI-H460 Cells

摘要 [目的]探讨沙立度胺(thalidomide,TD)及其对映体(S-thalidomide)S-TD对人非小细胞肺癌系NCI-H460细胞增殖的影响。[方法]体外培养NCI-H460细胞,采用MTT比色法测定不同质量浓度的TD、S-TD对NCI-H460细胞增殖的影响;采用流式细胞仪检测对细胞周期的影响;DAPI染色观察诱导细胞凋亡作用。[结果]在10.0~200.0μg/ml范围内,TD、S-TD作用于NCI-H460细胞24h可抑制细胞增殖、48h显著抑制细胞增殖,而在高浓度时(200.0μg/ml)S-TD的抑制作用弱于TD;TD、S-TD作用NCI-H460细胞24h、48h、72h,TD的抑制率分别为25.69%、41.92%、7.57%,S-TD为14.77%、33.24%、8.46%。200.0μg/mlTD和S-TD处理NCI-H460细胞48h后,G2/M期细胞比例显著增高,S期细胞比例下降。DAPI染色结果发现细胞数量明显减少,但未出现核固缩、核边集等凋亡形态学改变。[结论]TD和S-TD在一定浓度范围对NCI-H460细胞具有抗增殖作用,但S-TD的抗增殖活性稍弱于TD。抑制增殖作用可能与阻滞G2/M期细胞周期有关,DAPI染色结果未显示诱导细胞凋亡作用。
