七氟烷预处理对大鼠缺血再灌注肝脏TNF-α及ICAM-1表达的影响 被引量:1

摘要 目的:探讨肿瘤坏死因子(TNF-α)和细胞间粘附分子-1(ICAM-1)在七氟烷保护大鼠肝脏缺血再灌注损伤机制中的作用。方法:将40只SD成年雌性大鼠,随机平均分为4组(n=10):假手术组,缺血再灌注组,七氟烷组,七氟烷-缺血再灌注组。假手术组、七氟烷组作为对照;缺血再灌注组和七氟烷-缺血再灌注组阻断支配大鼠肝脏左叶和中叶的门静脉造成约70%肝脏缺血再灌注模型,缺血1h、再灌注2h后取材。测定大鼠血清ALT和A ST作为肝损害的标志被检测,光镜观察组织学病理改变,电镜下观察肝细胞的超微结构,采用免疫组织化学法检测肝组织TNF-α活性和ICAM-1水平。结果:七氟烷-缺血再灌注组,肝ALT、A ST酶升高受到显著抑制,电镜显示七氟烷-缺血再灌注组细胞损伤程度小于缺血再灌注组,七氟烷-缺血再灌注组TNF-α和ICAM-1表达低于缺血再灌注组。结论:七氟烷预处理能抑制肝缺血再灌注细胞的TNF-α和ICAM-1表达;TNF-α可能通过调控ICAM-1的表达在缺血再灌注损伤中发挥作用。 Objective:To investigate the role of the TNF-α and intercellular adhesion molecule-1(ICAM-1) in the sevoflurane against hepatic ischemia-reperfusion(IR) injury mechanism.Methods:40 adult female SD rats were randomly divided into 4 groups(n=10): Sham-operated group;Ischemia-reperfusion group;Sevoflurane group;Sevoflurane ischemia-reperfusion group.Sham-operated group,sevoflurane group served as controls;in ischemiareperfusion group and sevoflurane ischemia-reperfusion group,the hepatic hilar vessels distributing to the left and median lobes were clamped to induce partial hepatic ischemia(70%).Reperfusion was done 1 hour after hepatic ischemia.The animals were killed 2h after reperfusion,then the specimens of liver tissues and blood were obtained.ALT and AST in blood serum were detected as liver damage markers.The histological examination of liver tissues was done by light microscope,the ultrastructure of liver cells was observed by electron microscopy.The levels of TNF-α and ICAM-1 in the hepatic tissues were detected by immunohistochemistry.Results: In sevoflurane ischemia-reperfusion group,liver ALT,AST enzymes were significantly inhibited,electron microscopy showed that the degree of injury in sevoflurane-ischemia-reperfusion group was smaller than that in ischemia-reperfusion group;The expression of TNF-α and ICAM-1 in sevoflurane-ischemia-reperfusion group was slower than that in ischemia-reperfusion group.Conclusion:Sevoflurane preconditioning inhibits the expression of TNFα and ICAM-1 in hepatic ischemia-reperfusion.TNF-α may regulate the expression of ICAM-1 in ischemia-reperfusion injury.
