摘要 目的:建立实验性自身免疫性重症肌无力(EAMG)大鼠模型。方法:采用亲和层析技术从电鳗电器官中提取、纯化乙酰胆碱受体,以此免疫Lewis大鼠,观测其临床表现、乙酰胆碱受体抗体及重复神经电刺激等变化。结果:模型组大鼠出现肌无力症状,重复神经电刺激动作电位衰减率及乙酰胆碱受体抗体检测均为阳性,有统计学意义,对照组无明显改变。结论:乙酰胆碱受体主动免疫Lewis大鼠可成功诱导产生EAMG大鼠模型。 Objective:To set up a model of experimental autoimmune myasthenia gravis( EAMG) rats. Methods:Immunized Lewis rats by Acetylcholine receptors( AChR) to be extracted and purified from the electric organs of California Torpedo by affinity chromatography,observed rats' scales of clinical manifestation,repetitive nerve stimulation and determined anti-AChR antibodies. Results:The model group rats showed the typical evidence of EAMG including muscle weakness,a decremental response on repetitive nerve stimulation and increasing titers of anti-AChR antibodies,In control group,the rats immunized with placebo showed no findings of EAMG. Conclusion:AChR can induce EAMG by active immunity.
