摘要
目的:探讨姜黄素对经转化生长因子β(TGF-β1)诱导后人胚肺成纤维细胞一型胶原与c-jun,在人胚肺成纤维细胞中的表达的影响及其与特发性肺纤维化的关系。方法:细胞分为正常组、姜黄素组、模型组、治疗组采用采用RT-PCR,Western bloting法检测、I型胶原与c-jun在人胚肺成纤维细胞的变化。结果:治疗组与正常组、模型组相比,I型胶原与c-jun,表达明显减弱。结论:姜黄素可抑制人胚肺成纤维细胞的胶原沉积,在延缓肺纤维化形成中起一定作用,其作用机制可能是通过抑制核转录因子c-jun表达而实现的。
Objective:To discuss the preventing and treating effect of curcumin on pulmonary fibrosis(PF),this ex-periment has adopted MRC-5 induced by transforming growth factor 1(TGF-β1),to explore the effect and mecha-nisms of curcumin on PF from molecular level in vitro.Methods:The MRC-5 were divided into four groups :normal group;curcumin group;model group;treatment group.The change of I type collagen and c-jun expression were tested by RT-PCR and western blotting in MRC-5.Results:Compared with normal group and model group,it is easy to see that I type collagen and c-jun expression weaken obviously in treatment group.Conclusions:Curcumin can inhibit the MRC-5’s collagen deposition which may have important role in the delaying lung fibrosis forming progress.Its mecha-nism may be through the inhibiting nuclear transcription factor:c-jun expression.
