中药复方“松友饮”含药血清体外抑制高转移人肝癌MHCC97H细胞的实验研究

The Experimental Research on the in Vitro Inhibitory Effects of Medicated Serum with Chinese Herbal Compound "Songyou Yin" on Human Hepatocellular Carcinoma MHCC97H Cells with High Metastatic Potential

目的:研究中药复方"松友饮"含药血清对体外培养的人肝癌MHCC97H细胞增殖、侵袭潜能的影响。方法:以MHCC97H细胞构建裸鼠原位移植瘤模型,84只成模裸鼠随机平均分成对照组和"松友饮"组,肝脏接种肿瘤24 h内开始1次/天灌胃给药。在第1、4、7、14、21、28、35次灌胃结束后2h,每组分别处死6只裸鼠收集血样。应用含药血清体外干预MHCC97H细胞,检测细胞增殖、侵袭潜能变化。结果:"松友饮"灌胃给药1、4、7天的荷HCC裸鼠含药血清体外抑制HCC作用逐渐增强,给药7、14、21、28、35天的含药血清抑制作用无统计学差异。灌胃7天的"松友饮"含药血清抑制HCC增殖的作用与中剂量卡培他滨作用近似,抑瘤率分别为37.4%、41.1%。给药7天的含药血清抑制HCC侵袭潜能达到最强,"松友饮"组穿膜细胞数明显少于对照组,为26.0±3.6和37.3±2.8。结论:给药7天的含药血清达到稳态血药浓度,对MHCC97H细胞的体外抑制作用达到最强。

Objective：To investigate the in vitro effects of medicated serum with Chinese herbal compound ＂Songyou Yin＂ on human hepatocellular carcinoma（HCC） MHCC97H cells with high metastatic potential.Methods：Metastatic human HCC nude mice models were established by orthotopic implantation with human HCC cell line MHCC97H.Eighty four nude mice bearing orthotopic xenografts were randomized into control group and ＂ Songyou Yin＂ group.Daily intra-gastric administration of ＂ Songyou Yin＂ was given 24 hours after orthotopic implantation.Two hours after the 1st,4th,7th,14th,21st,28th,35th intragastric administration,mice were sacrificed by cervical dislocation to collect serum sam-ple,which were used to intervene the MHCC97H cells for examination of proliferation and invasion of MHCC97H cells.Results：The in vitro inhibitory effects of medicated serum on MHCC97H cells were stepwise increased after the 1st,4th,7thintragastric administration of ＂ Songyou Yin＂,but the difference in the effects of medicated serum of the 7th,14th,21st,28th,35th administration was minor.The inhibitory effects of medicated serum of the 7th administration on HCC proliferation was similar with that of capecitabine（0.4 mg/L）,the inhibitive rates were 37.4% and 41.1% respectively.The invaded cells in control group（37.3 ± 2.8） were significantly more than those treated by medicated serum of the 7th administration in ＂ Songyou Yin＂ group（26.0 ± 3.6）.Conclusion：The medicated serum concentration of ＂ Songyou Yin＂ after the 7th intragastric administration was stable with the inhibitoriest in vitro effects on MHCC97H cells
