桑椹三仙胶对慢性特发性血小板减少性紫癜患者共刺激分子CD_(80)和CD_(86)表达的影响

Effects of Mulberry Sanxian Gum on the Expressions of CD_(80) and CD_(86) mRNA in the Eatients with Chronic Idiopathic Thrombocytopenic Purpura

目的:观察桑椹三仙胶对慢性特发性血小板减少性紫癜病人外周血单个核细胞CD80和CD86的mR-NA的影响。方法:采用RT-PCR法测定桑椹三仙胶治疗前后CITP病人及正常对照组外周血单个核细胞CD80和CD86的mRNA的表达。结果:CITP组CD80和CD86mRNA表达有相同倾向,即均高于正常组,治疗后均有所下降。CITP组CD80、CD86mRNA表达较正常对照组明显增高,具有显著性差异(P<0.01);桑椹三仙胶治疗后CD80、CD86mRNA表达较治疗前明显降低,差异显著(P<0.01),但未达到正常水平,与正常组比较仍有显著性差异(P<0.01)。结论:CITP患者外周血单个核细胞存在共刺激分子CD80、CD86的过度表达。桑椹三仙胶可能通过纠正共刺激分子异常表达调节免疫状态,从而达到缓解甚至治愈CITP的目的。

Objective：To observe the effects of Mulberry Sanxian Gum（MSG） on the expressions of CD80 and CD86 mRNA in peripheral blood mononuclear cells from patients with chronic idiopathic thrombocytopenic purpura（CITP）.Methods：RT-PCR method was used to measure the levels of CD80 and CD86 mRNA in peripheral blood mononuclear cells in patients with CITP and normal control subjects before and after MSG treatment.Results：The expressions of CD80 and CD86 mRNA in CITP group showed the same trends that the values were higher than normal controls and reduced after treatments.The levels of CD80 and CD86 mRNA in CITP group were significantly higher than that in normal control group（P 〈0.01）.After MSG treatments,the expressions of CD80 and CD86 mRNA were reduced remarkably compared with that before treatments.But their levels did not return to normal and the difference was significant compared with normal control group（P 〈 0.01）.Conclusion：There were overexpressions of costimulatory molecules CD80 and CD86 in periph-eral blood mononuclear cells from patients with CITP.MSG may work through modifying the abnormal expression of cos-timulatory molecules,whereby regulating immune status so as to achieve remission or even cure CITP
