摘要
目的通过移植骨髓间充质于细胞(BMSCs)联合甲基强的松龙(MP),观察其对百草枯(PQ)中毒急性肺损伤的防治效果。方法54只雌性受体大鼠随机分为5组:PQ组、BMSCs组、MP组、BMSCs+MP组和正常对照组,观察7d内各组大鼠的肺湿干比、血浆中IL-18、TNF-α、MDA、SOD的水平变化。结果与PQ组相比,BMSCs组和MP组能明显降低肺湿干比(P≤0.01)和炎症介质IL-1β、TNF—α的含量(P≤0.01),降低血浆中MDA水平(P≤0.01)和升高SOD水平(P≤0.01)。但BMSCs+MP组的上述指标均显著优于BMSCs组和MP组(P≤0.01)。结论BMSCs与MP的联合应用,既起到抑制炎症反应、减少脂质过氧化产物的协同作用,又促进移植BMSCs的体内存活,增强了移植效果。
Objective To investigate the protective effects of bone marrow mesenchymal stem cells (BMSCs) and methylprednisolone (MP) on Paraquat(PQ)-induced acute lung injury in rats. Methods 54 female recipient SD rats were divided randomly into five groups- PQ group, BMSCs group, MP group, BMSCs+MP group and normal control group. The lung wet/dry weight ratios were calculated during 7 days after injections in each grouP. At the same time, the levels of Interleukin-1 β (IL-1β), tumor necrosis faetor-α (TNF-α), malondialdehyde '(MDA), superoxide dismutase (SOD) in plasma were detected. Results Compared with PQ group, the lung wet/dry weight ratios were down obviously in BMSCs group and MP group, at the same time, the levels of IL-1β, TNF-α, and MDA in plasma were decreased markedly, and the activities of SOD in plasma were increased significantly in the both groups. However, the detective results in BMSCs + MP group were significantly better than those in BMSCs group and MP group. Conclusion BMSCs and MP applications can not only restrain inflammatory reaction and reduce lipid peroxide, but also enhance the effect of BMSCs transplantation.
