多聚赖氨酸包被球囊导管转移基因表达的时相特征 被引量:5

摘要 目的评估多聚赖氨酸包被球囊导管-重组腺病毒/LacZ(Ad5/LacZ)局部基因转移系统的基因转移效率与表达时间。方法利用该系统,在体将Ad5/LacZ导入兔腹主动脉下段。然后于转基因后不同的时间点对转基因表达进行了定性(X-Gal染色)及定量(β-Gal活性)测定。结果基因转移后7天的表达最高。3天与14天的次之,21天时表达显著下降。28天时几乎测不到转基因的表达。结论该基因转移系统能有效地实现直接、在体的基因转移。腺病毒介导的基因表达时间是短暂的。其表达峰值约在7天左右。 Objective The expression and time course of the transgene induced by polylysine coated balloon catheter system carring Ad5/LacZ were evaluated in this study Methods With the local gene delivery system,the reporter gene LacZ carried in Ad5 Vector were transferred into the low parts of the rabbit abdominal aortic arteries,and after the procedures,the expression and time course of the transgene were detected with Qualitatively (X Gal staining) and quantitatively (β Gal activity) assay at different time points Results The expression of the gene increased progressively reaching the peak at day 7 It declined significantly at day 21 and almost were not detectable at day 28 The gene expression were similar at day 3 and 14 after the gene transfer Conclusion The system could effectively directly achieve gene transfer in vivo Transgene expression mediated by the recombinant adenovirus were transient
