摘要
目的:研究乙型肝炎病毒基因型与核心启动子及前C区突变的关系。方法:采用基因测序方法测定138例乙型肝炎病毒S基因序列,并比对分析确定基因型;同时采用基因芯片方法检测了核心启动子及前C区突变情况。统计分析基因型与突变的关系。结果:核心启动子T1762/A1764在C基因型中突变明显高于B基因型(P<0.01),而1814及前C区1896位点突变在B和C基因型中差异无显著性(P>0.05)。B基因型与C基因型在HBeAg阳性和阴性患者中的分布差异无显著性(P>0.05)。结论:乙型肝炎病毒C基因型更容易发生T1762/A1764双突变。
Objective:To probe the relationship between HBV genotype and mutations in pre-core and basic core promoter regions.Methods: The S genes of 138 samples with HBV-DNA were sequenced and the genotype was identified by BLAST,and the microarray was applied to detect the mutations in pre-core and basic core promoter regions.The relationship of the genotype and mutations was evaluated statistically.Results: The T1762/A1764 mutation rate in C genotype group was significantly higher than that in B genotype group(P0.01);but there was no significant difference in 1814 and 1896 site mutation between B and C genotype group.The HBV genotype was also not related with the status of HBeAg.Conclusion: The T1762/A1764 double-mutation in basic core promoter regions was more common in HBV C genotype than B genotype.
