摘要
目的研究健康受试者口服异丙肌苷片(1.0 g)的药动学特征,并考察高蛋白高脂肪食物对其药动学的影响。方法采用随机双周期交叉设计,12名健康受试者(男女各半),随机分为2组,I组空腹、II组进食后均分别口服异丙肌苷片(1.0 g),服药后0~24 h内间隔采集血样,停药1 wk后进行交叉试验。采用液相色谱-串联质谱法(LC-MS/MS)和高效液相色谱法(HPLC)分别测定血浆中N,N-二甲氨基-2-丙醇(Dip)和4-乙酰氨基苯甲酸(PACBA)的浓度。结果空腹和进食后单剂量口服异丙肌苷片,Dip的ρmax分别为(1.6±s 0.4)和(1.7±0.6)mg.L-1;tmax分别为(1.4±0.8)和(1.8±1.2)h;t1/2分别为(3.7±0.7)和(3.6±0.7)h;MRT分别为(5.6±1.0)和(5.8±1.1)h;AUC0~24分别为(7.9±1.8)和(8.0±1.9)mg.h.L-1。PACBA的ρmax分别为(9±4)和(3.4±1.7)mg.L-1;tmax分别为(0.59±0.20)和(1.5±1.1)h;t1/2分别为(1.02±0.24)和(1.2±0.4)h;MRT分别为(1.46±0.29)和(2.4±0.7)h;AUC0~24分别为(10±4)和(7.6±1.8)mg.h.L-1。结论异丙肌苷血浆样品LC-MS/MS、HPLC测定法专属、灵敏、准确,适用于异丙肌苷的临床药动学研究。进食高脂肪、高蛋白食物对Dip的药动学没有显著影响,但对PACBA的吸收影响明显。
AIM To study the pharmacokinetics of inosiplex tablets(1.0 g) in healthy volunteers and the effect of high protein and fat diet on it.METHODS The investigation was carried out according to an open,randomized,2-period crossover design with a 1-week washout interval.Twelve volunteers(6 men and 6 women) were randomly divided into 2 groups.Inosiplex tablets(1.0 g) were administrated in the fasting or fed state.Blood samples were collected in 24 h at intervals.The plasma concentrations of N,N-dimethylamino-2 propanol (Dip) and p-acetaminobenzoic acid(PACBA) were measured by fully validated LC-MS/MS and HPLC method,respectively.RESULTS The major pharmacokinetic parameters of the single-dose study in fasting and fed states were as follows: for Dip,ρmax were(1.6 ± s 0.4) vs.(1.7 ± 0.6) mg.L-1,tmax were(1.4 ± 0.8) vs.(1.8 ± 1.2) h,t1/2 were(3.7 ± 0.7) vs.(3.6 ± 0.7) h,MRT were(5.6 ± 1.0) vs.(5.8 ± 1.1) h,AUC0-24 were (7.9 ± 1.8) vs.(8.0 ± 1.9) mg.h.L-1,respectively.In the case of PACBA,ρmax were(9 ± 4) vs.(3.4 ±1.7) mg.L-1,tmax were(0.59 ± 0.20) vs.(1.5 ± 1.1) h,t1 /2 were(1.02 ± 0.24) vs.(1.2 ± 0.4) h,MRT were(1.46 ± 0.29) vs.(2.4 ± 0.7) h,AUC0-24 were(10 ± 4) vs.(7.6 ± 1.8) mg.h.L-1,respectively.CONCLUSION The established LC-MS/MS and HPLC methods for the determination of Dip and PACBA are specific,sensitive,accurate,and suitable for clinical investigation of inosiplex pharmacokinetics.High protein and fat diet has no obvious influence on the pharmacokinetics of Dip,but affects the absorption of PACBA.
