白芍总糖苷对RA大鼠足爪组织中NF-κB/p65蛋白表达的抑制作用

Inhibitory effect of total glucosides of paeonia on the NF-κB/p65 protein expression in paws of RA rats

目的:探讨白芍总糖苷(TGP)对类风湿性关节炎(RA)大鼠足爪组织中核转录因子NF-κB/p65蛋白表达的抑制作用和对血清中炎性细胞因子IL-1β、TNF-α、IL-10等水平的影响。方法:建立Ⅱ型胶原诱导的RA大鼠模型。用免疫组化染色法检测足爪组织中NF-κB/p65蛋白的表达。用ELISA法检测RA大鼠血清中IL-1β、TNF-α和IL-10的含量。结果:TGP能以剂量依赖性地下调NF-κB/p65蛋白的表达,降低RA大鼠血清中TNF-α、IL-1β的水平。结论:TGP对RA大鼠炎症的抑制作用,可能与其下调NF-κB/p65蛋白的表达,抑制促炎性细胞因子TNF-α、IL-1β的产生有关。

AIM： To explore effects of total glucosides of paeonia（TGP）on the NF-κB/p65 protein expression in paws of rheumatoid arthritis（RA） rats,and the contents of serum TNF-α,IL-1β and IL-10 in RA rats serum.METHODS： Type Ⅱcollagen-induced arthritis model in SD rats was established.The expression of NF-κB/p65 proteins in rat,s paw tissues was detected by immunohistochemical staining.The contents of serum TNF-α,IL-1β and IL-10 in RA rats were detected by ELISA.RESULTS： TGP could down-regulate the expression of NF-κB/ p65 protein and contents of TNF-α,IL-1β and IL-10 in RA rats.CONCLUSION： The inhibitory effect of TGP on antiinflammation maybe related to decreasing the expression of NF-κB/p65 protein and suppressing the production of TNF-α and IL-1β in RA rats.
