暖心胶囊对心力衰竭模型大鼠的心肌细胞及P53蛋白表达的影响

Effects of "Nuanxin Capsule" on cardiocytes and expression of P53 protein in heart failure rats

目的探讨暖心胶囊对心力衰竭(心衰)模型大鼠的心肌细胞及P53蛋白表达的影响。方法 40只SD大鼠随机分为空白组、模型组、暖心胶囊低剂量组、暖心胶囊高剂量组、地高辛组。除空白对照组外,其余各组均用狭窄腹主动脉方法造模,成功模型灌胃给药,测量心肌重量,观察心肌病理改变,运用免疫组织化学染色法检测P53蛋白。结果与空白组比较,模型组心脏比重明显增加,P53蛋白表达阳性率显著升高,差异有统计学意义(P<0.05);与模型组比较,暖心胶囊低剂量组、暖心胶囊高剂量组、地高辛组能明显降低心脏比重(P<0.05),明显改善心衰大鼠的心肌结构,减少P53表达(P<0.05)。结论暖心胶囊能明显改善心衰大鼠的心肌结构,逆转心肌重构,减少P53表达,拮抗心肌细胞的凋亡。

Objective To explore the effects of ＂Nuanxin Capsule＂ on the cardiocytes and the expression of P53 protein in heart failure rats. Methods Forty SD rats were randomly classified into blank group, model group, Nuanxin Capsule Group（low dosage, high dosage） and digoxin group. Models were made by narrowing the abdominal aorta in groups other than the blank group. Intragastric administration was given to the successful models. Then the cardiac muscle was weighed and the pathological changes and P53 protein were assayed with immunohistochemical stain method. Results In Nuanxin Capsule low-and high-dosage groups and digoxin group, the heart specific gravity was lowered（P0.05）, the cardiac muscle structure was improved and the expression of P53 was decreased（P0.05）. Conclusion ＂Nuanxin Capsule＂ can remarkably improve the cardiac muscle structure, reverse the reconstruction of cardiac muscle, lessen the expression of P53 and relieve the apoptosis of the cardiocytes.
