大麦黄矮病毒PAV河南分离物通读蛋白ORF5基因的分子特征分析

Analyses of molecular characteristics on the ORF5 gene of read through protein of barley yellow dwarf viruses-PAV isolated from Henan

摘要 利用RT-PCR从河南各地采集的110份小麦病样上鉴定出28个大麦黄矮病毒(Barley yellow dwarf viruse,BYDV)PAV分离物,并获得了这28个分离物的通读蛋白(Read through protein,RTP)ORF5基因全长.通过氨基酸序列比对可知,河南分离物RTP-ORF5基因变异普遍较大,28个BYDV-PAV河南分离物RTP-ORF5基因之间的核苷酸序列一致性为75.9%~100%,氨基酸序列一致性为79.4%~99.8%,核苷酸和氨基酸的一致性变异都较大.与国内外报道的16个BYDV-PAV通读蛋白基因相比对,其核苷酸序列一致性为72.2%~99%,由此推导的氨基酸序列一致性为77%~99.6%.系统进化分析显示河南地区分离物在4个内组群中都有分布.这些数据说明河南的大麦黄矮病毒PAV多样性丰富. 28 isolates of BYDV-PAV were detected by RT-PCR from 110 infected wheat plants which were collected from Henan Province.The ORF5 genes of Read through protein(RTP)of these isolates were sequenced and analyzed.The identities of ORF5 gene between the 28 isolates of BYDV-PAV were 75.9%~100% on nucleotide acids and 79.4%~99.8% on amino acids respectively.When the obtained sequence data were compared with previously reported sequences of 16 BYDV-PAV isolates home and abroad,the results showed that the identities of ORF5 gene were 72.2%~99% on nucleotide acids and 77%~99.6% on amino acids respectively.The phylogenetic analysis of BYDV-PAV isolates described the evolution relationship among the isolates from worldwide including China,the four subgroups were found in the phylogenetic tree,the Henan isolates distributed in four subgroups.While,the isolates colleted from Henan Province were not clustered by geographical origins or collecting time.The data suggested that BYDV-PAV isolates in Henan may be rich in biodiversity.
