摘要
研究了提取大豆蛋白质工艺中pH值对蛋白质提取率及得率的简单效应,结果表明:①在料水比1∶10、酸沉pH值4.3、离心速度4 000 r/min、碱溶pH值9.5、碱溶时间为45 min的条件下,蛋白质平均提取率最高为66.49%。在pH值7.5~9.0之间,碱溶pH值对蛋白质提取率的简单效应为2.88%;在pH值9.0~10.0之间,简单效应下降直至为负值。方差分析表明,各处理间蛋白质提取率差异极显著;新复极差测验表明,pH值7.5~9.0之间时,各处理的蛋白质提取率差异极显著,pH值9.0~10.0之间时,差异不显著。②同样条件下,蛋白质平均得率最高为37.23%。在pH值7.5~9.0之间,碱溶pH值对蛋白质得率的简单效应为2.33%;在pH值9.0~10.0之间,简单效应下降直至为负值。方差分析表明,各处理间蛋白质得率达显著水平;新复极差测验表明,pH值7.5~8.5之间时,各处理间的蛋白质得率差异显著,pH值8.5~10.0之间时,差异不显著。③在不考虑其他因素时,为了取得碱溶pH值的最大简单效应,宜选择9.0作为碱溶pH值参数。
The simple effect of alkali soluble pH value on the extraction rate of soybean protein(ERSP)and yield rate of soybean protein(YRSP) was studied.The results showed that:①When the ratio of material to water was 1∶10,acid precipitation pH was 4.3,centrifugal speed was 4 000 r/min,alkali-soluble pH value was 9.5 and the alkali-soluble was 45 min,the highest mean of ERSP was 66.49%.Among pH 7.5~9.0,the simple effect of alkali soluble pH value on the ERSP was 2.88%.However,among pH 9.0~10.0,the simple effect reduced to a minus value.Analysis of variance showed discrepancy of ERSP among different treatments reached very notable level;The shortest significant range showed,when pH was 7.5~9.0,discrepancy of ERSP reached very notable level,when pH was 9.0~10.0,it was no difference.②When in the same conditions,the highest mean of YRSP was 37.20%.Among pH 7.5~9.0,the simple effect of alkali soluble pH value on the YRSP was 2.33%.The simple effect also reduced among pH 9.0~10.0.Analysis of variance showed discrepancy of ERSP among different treatments reached notabale level;The shortest significant range showed when pH was 7.5~8.5,discrepancy of YRSP was significant difference,when pH was 8.5~10.0,it was no difference.③In order to obtain the biggest simple effect of alkali soluble pH value,pH 9.0 was suggested in the extraction process of soybean protein.
