摘要
目的研究α干扰素(αIFN)对膀胱癌多药耐药性的逆转作用。方法应用MTT法检测阿霉素(ADM)的细胞杀伤作用。荧光法检测细胞内阿霉素浓度和斑点杂交法检测mdr1mRNA相对含量。结果500IU/mlαIFN能明显增强阿霉素对耐药细胞BIU87/ADM的杀伤作用,但不能增强阿霉素对敏感细胞BIU87的杀伤作用;αIFN能增加阿霉素在BIU87/ADM细胞内的积聚,但不改变mdr1mRNA在细胞内表达量。结论αIFN对膀胱癌细胞多药耐药性具有逆转作用,其机制与αIFN增加细胞内抗癌药物浓度有关,与增加mdr1mRNA表达量无关。
Objective To study the reversal of multidrug resistance by α IFN in bladder cancer cell line. Methods The effect of recombinant human α interferon (αIFN) on the cytotoxicity of adriamycin(ADM) in human bladder cancer cell line BIU 87 and its multidrug resistant counterpart BIU 87/ADM were studied by MTT assay. ADM accumulation and level of mdr1 mRNA in above cell lines were also detected by spectrofluometric and dot blot analysis respectively. Results α IFN (500IU/ml) increases the cytotoxicity effect and the accumulation of ADM in BIU 87/ADM cell line, but had no effect on the cytotoxicity of ADM to BIU 87 cell line, the level of mdr1 mRNA being unchanged. Conclusions α IFN has significant effects on the reversal of multidrug resistance of human bladder cancer cell line.
