摘要
目的通过静脉注射氟尿嘧啶建立稳定有效的化疗性静脉炎小鼠模型。方法 42只昆明小鼠按照给药浓度随机分为7.5 mg/mL、10.0 mg/mL、12.5 mg/mL,15.0 mg/mL、17.5 mg/mL和22.5 mg/mL共6个实验组和1个对照组。实验组小鼠分别从左侧鼠尾静脉注射0.4 mL不同浓度的氟尿嘧啶溶液,对照组注射0.4mL的生理盐水。给药后第3 d对化疗性静脉炎表现进行分级评价后处死。制作石蜡切片并进行镜下分级。结果随注射氟尿嘧啶剂量的增加,小鼠静脉炎发生率也逐渐增高,350 mg/kg(17.5 mg/mL组)以上剂量组动物出现中毒死亡。300 mg/kg(15.0 mg/mL)剂量组化疗性静脉炎发生率达100%,无动物死亡,出现典型的静脉炎症状,对照组未出现类似变化。结论通过小鼠尾静脉注射300 mg/kg~350 mg/kg剂量范围的氟尿嘧啶可成功建立稳定有效的化疗性静脉炎小鼠模型。
Objective To establish a stable and effective chemotherapy induced phlebitis(CIP) model in mice by intravenous injection of Fluorouracil.Methods Forty two KM mice were randomly divided into 6 treatment groups and 1 control group.The treatment groups were administrated with Fluorouracil at doses of 7.5 mg/mL,10.0 mg/mL,12.5 mg/mL,15.0 mg/mL,17.5 mg/mL,and 22.5 mg/mL respectively,and each mouse received Fluorouracil by injection of 0.4 mL volume into the left caudal vein.The control received the same volume of saline instead.All the mice were sacrificed at the third day after treatments.The degree of lesions was evaluated and pathological changes were examined under a microscope.Results The incidence rate of CIP showed a dependence on the dose of Fluorouracil.The mice dead when the dose was higher than 350 mg/kg(17.5 mg/mL group).The incidence rate of CIP was 100% and the typical symptom of CIP was observed when the treatment groups received injection of Fluorouracil at 350mg/kg(15.0 mg/mL group),with all mice surviving.But those changes were not observed in the control group.Conclusion The stable and effective mouse model of CIP can be established by caudal vein injection of Fluorouracil at doses from 300 mg/kg to 350 mg/kg.
