OGF、紫杉醇对乳腺癌MCF-7细胞增殖的影响及机制探讨 被引量:4

摘要 目的观察阿片生长因子(OGF)、紫杉醇对人乳腺癌MCF-7细胞增殖的影响,并探讨其机制。方法取对数生长期的MCF-7细胞,随机分为A、B、C、D、E、F、G组,分别用OGF、紫杉醇、OGF+紫杉醇、OGF受体抑制剂naloxone、OGF+naloxone、紫杉醇+naloxone、OGF+紫杉醇+naloxone处理,OGF浓度为10-7M,紫杉醇和naloxone浓度为10-6M,每组均设空白对照。采用MTT法测算各组MCF-7细胞增殖抑制率。结果 A、B、C、D、E、F、G组细胞增殖抑制率分别为25.94%±2.96%、46.98%±5.75%、55.45%±6.97%、2.77%±0.45%、6.38%±1.47%、46.42%±5.32%、47.09%±6.53%。MCF-7细胞增殖抑制率C组高于A、B组,E组低于A组,G组低于C组,P均<0.05;B组与F组、D组与E组相比P均>0.05。结论 OGF、紫杉醇均可抑制乳腺癌MCF-7细胞增殖,二者联用具有协同作用。OGF对MCF-7细胞增殖的抑制作用是OGFR介导的,紫杉醇对MCF-7细胞增殖的抑制作用与OG-FR无关。, Objective To observe the effect of combination of OGF and taxol on human breast carcinoma MCF-7 cells proliferation,and explore its mechanism.Methods MCF-7 cells were divided into A,B,C,D,E,F,G group,and were cultued with OGF,Taxol,OGF+Taxol,naloxone,OGF+naloxone,Taxol+naloxone,OGF+Taxol+naloxone.MTT essay was used to detect the MCF-7 cell proliferation inhibition rate in each group.Results The cell proliferation inhibition rate in A,B,C,D,E,F,G group was 25.94%±2.96%、46.98%±5.75%、55.45%±6.97%、2.77%±0.45%、6.38%±1.47%、46.42%±5.32%、47.09%±6.53%,respectively.Conclusions OGF and taxol can inhibit MCF-7 cell proliferation,respectively.The MCF-7 cell proliferation inhibition effect of OGF is nediated by OGFR,but taxol is not.
