摘要
目的观察沙芥对高脂饮食大鼠有无预防高血脂的作用及对血管内皮功能的影响。方法 64只Wistar大鼠,随机分为空白对照组、高脂模型组、血脂康药物组和沙芥高、中、低浓度组,除空白对照组给予普通饲料外,其余各组均饲喂高脂饲料,同时血脂康组和沙芥组,分别以血脂康混悬液和高、中、低浓度沙芥灌胃,空白、模型组灌等容积蒸馏水,每天一次,连续8周后处死。检测大鼠血清的总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C),血浆内皮素(ET)、一氧化氮(NO)、降钙素基因相关肽(CGRP)水平。结果高脂组大鼠较空白组TC、TG、LDL-C、ET显著升高,HDL-C、NO、CGRP水平显著降低,提示模型组高脂血症模型已形成,并存在血管内皮损伤。各沙芥组均较高脂模型组大鼠的TC显著降低;高浓度沙芥组较模型组大鼠的LDL-C、ET显著降低,NO显著升高;中浓度PCG组较模型组NO显著升高。结论沙芥对高脂饮食的大鼠有一定的预防高脂血症的作用,并有一定的保护血管内皮功能的作用。
Objective To examine the potential role of Pugionium cornutum(L.)Gaertn(PCG)in prevention of hyperlipidemia in rats fed with high fat diet as well as its effect on function of vascular endothelia.Methods Sixty four Wistar rats with half males and half females were randomly divided into six groups including normal control group,fatty liver model group,Xuezhikang-treated group,high dosage PCG-treated group,medium dosage PCG-treated group and low dosage PCG-treated group.Except rats in control group fed with normal diet,all rats in other groups were fed with high fat diet.Rats in Xuezhikang-treated group and three PCG-treated groups were subjected to administration with either Xuezhikang suspension(0.3 g/kg)or PCG(10.38 g/kg as high dosage,5.19 g/kg as medium dosage and 2.60 g/kg as low dosage)respectively while rats in control group and fatty liver model group were subjected to administration with equal volume of distilled water.After stomach perfusion was performed in each rat once a day for eight consecutive weeks,all rats were decapitated and examined for serum indices of following category:total cholesterol(TC),triglyceride(TG),high density lipoprotein-cholesterol(HDL-C),low density lipoprotein-cholesterol(LDL-C),plasma endothelin(ET),nitric oxide(NO)and calcitonin gene-related peptide(CGRP).Results In comparison with control group,rats in fatty liver model group were detected with a marked increase in levels of TC,TG,LDL-C,and ET and a marked decrease in levels of HDL-C,NO and CGRP indicating the successful formation of hyperlipidemia model as well as the existence of endothelial injury.Rats in all PCG-treated groups exhibited markedly decreased TC level in comparing with that of those in fatty liver model group.In comparison with fatty liver model group,rats in high dosage PCG-treated group exhibited significantly reduced LDL-C and ET levels and also markedly increased NO level,the latter of which was also detected in rats of medium dosage PCG-treated group.Conclusion PCG exerted certain effect in prevention of hyperlipidemia in rats fed with high fat diet and in preservation of vascular endothelial function.
