摘要
变性梯度凝胶电泳(PCR-DGGE)具有可靠性强、重复性好、方便快捷等优点,目前被广泛地应用于环境微型生物群落多样性和动态分析。本文对PCR-DGGE技术的基本原理和流程,图谱的优化过程和图谱分析方法,及其在环境微型生物生态学中的应用进行了综述,并对该技术自身存在的局限性和应用前景进行了评价。
PCR-denaturing gradient gel electrophoresis(PCR-DGGE) has the advantages of high reliability,good repetition,and easy operation.Currently,it has been widely used in the study of diversity and dynamics of microbial communities.This paper introduces the principle and procedure of PCR-DGGE,discusses the optimization and statistical analysis of PCR-DGGE profile,reviews its application in the study of environmental microbial ecology,and analyzes its limitations and potential application.
