阻断NF-κB通路对梗阻性黄疸大鼠肝细胞凋亡的影响

摘要 目的:探讨梗阻性黄疸时肝组织核因子-κB(nuclear factor kappa B,NF-κB)的表达与细胞凋亡的变化,并观察NF-κB特异性抑制剂吡咯烷二硫代氨基甲酸酯(PDTC)对肝脏损伤的保护作用。方法:将雄性SD大鼠72只随机分成胆总管结扎组(BDL组)、胆总管结扎+PDTC干预组(PDTC组)、假手术组(Sham组)。分别于术后第3天、第7天、第14天处死大鼠,观察肝组织细胞凋亡和NF-κB p65蛋白的表达。结果:BDL组和PDTC组NF-κB p65表达及细胞凋亡数明显增加,但在PDTC组明显低于BDL组。结论:NF-κB的活化可能是梗阻性黄疸时介导肝脏功能损害的重要因素。 Objective To investigate the expressions of nuclear factor kappa B(NF-κB)and apoptosis in liver of obstructive jaundice rats and the protective effect of NF-κB inhibitor(PDTC).Method Seventy-two male Sprague-Dawley rats were randomly divided into three groups:bile duct-ligation(BDL)group,bile duct-ligation plus PDTC(PDTC)group and sham-operation(Sham)group.The rats were sacrificed at 3,7 and 14 days after operation.Hepatic NF-κB activities and cell apoptosis were examined.Results Hepatic expression of NF-κB and apoptotic index in PDTC group were obviously lower than those in BDL group.Conclusion NF-κB may be one of the most important factors in liver damage induced by obstructive jaundice.
