摘要
利用荧光定量PCR技术对9个葡萄花发育相关基因在藤稔葡萄夏芽成花过程中的表达进行分析。所研究的基因包括:Vitis vinifera AGAMOUS(VvAG)、Vitis vinifera APETALA3(VvAP3)、Vitis vinifera FLOWERING LOCUSC(VvFLC)、Vitis vinifera FRUITFUL(VvFUL)、Vitis vinifera FLOWERING LOCUS T(VvFT)、Vitis vinifera APETALA2(VvAP2)和Vitis vinifera SUPPRESSOR OF OVER EXPRESSION OF CO1(VvSOC1)、Vitis vinifera APETALA1(VvAP1)以及Vitis vinifera LEAFY(VvLFY)。结果表明,经摘心处理的夏芽能够进行花芽分化及花芽形成,而未经处理的对照组夏芽未形成花芽;9个基因在处理与对照夏芽及发育枝茎尖中的表达存在一定的差异,其在处理样品中的表达与拟南芥中同源基因的表达基本相符。该研究对于深入了解葡萄花发育的分子机理,相关基因在转基因中的利用以及其表达的合理调控等具有重要的理论指导意义。
In this research, the quantitative real-time RT-PCR was employed to analyze the expression of nine important floral genes in grapevine cv. Fujiminori, i.e. Vitis vinifera A GAMOUS (VvA G), Vitis vinifera APETALA 3 (VvAP3), Vitis vinifera FLOWERING LOCUS C (VvFLC), Vitis vinifera FRUITFUL (VvFUL), Vitis vinifera FLOWERING LOCUS T (VvFT), Vitis vinifera APETALA2 (VvAP2), Vitis vinifera SUPPRESSOR OF OVER EXPRESSION OF CO 1 (VvSOC1), Vitis v inifera A PETA LA 1 (VvA P1) and Vitis vinifera LEA bY(VvLFY). The result showed that the summer buds on the shoots cut back could sprout much earlier and initiate flower bud differentiation and development, but the summer buds of control could not form floral bud and inflorescence. There were differences in the expression patterns of the nine genes between the treatments and control, and the expression of the genes in treated bud and shoot samples was basically the same as that of their homologous genes in A rcd)idopsis. This work could provide some information for further understanding of the molecular mechanism of flower development in grapevine, for the utilization of the related genes in genetic engineering, and for some positive control on their expression.
