摘要
缺氧诱导因子抑制因子(FIH)是体内调节缺氧诱导因子1(HIF-1)转录活性的主要因子之一。近年FIH研究中最引人注意的是一组含有锚蛋白重复序列结构域的底物蛋白质,这组蛋白普遍存在并参与多种生理功能,其含多个FIH羟化位点,但具有不完全羟化及优势底物的特征,又与FIH、HIF-1之间存在错综复杂的关系。
Factor inhibiting HIF-1( FIH) is one of the key components that control the activity of the master transcriptional regulator,hypoxia inducible factor-1( HIF-1) . Recently,an alternative class of FIH substrates which contain ankyrin repeat domain( ARD) is the focus of intense investigation. These proteins are ubiquitous and participate in many physiological processes. ARD substrates frequently contain multiple FIH hydroxylation sites and the hydroxylation is incomplete. Furthermore,ARD containing proteins have " preferred" substrates for FIH and also have complicated cross-talk with HIF and HIF-1 signaling.
