摘要
目的探讨17β-雌二醇(E2)和富含半胱氨酸蛋白61(Cyr61)影响人绒毛膜癌细胞系JEG-3细胞增殖的机制。方法定量PCR检测Cyr61和microRNA 195(miR-195)的表达,羧基荧光素乙酰乙酸琥珀酰亚胺酯(CFSE)染色法检测细胞增殖。结果 E2能够提高Cyr61 mRNA的表达;Cyr61能够降低miR-195的表达和促进JEG-3的细胞增殖。结论 E2通过增强Cyr61和降低miR-195的表达影响JEG-3细胞的增殖,可能为探讨子痫前期的发病机制提供线索。
Objective To investigate the mechanisms that 17β-estradiol(E2) and cysteine-rich 61(Cyr61) affect cell proliferation of human choriocarcinoma cell lines JEG-3 by regulating the expression of miR-195.Methods Cyr61 and miR-195 expression were determined by quantitative PCR.The cell proliferation was measured by carboxyfluorescein diacetate succinimidyl ester(CFSE).Results E2 increased Cyr61 mRNA level in JEG-3 cells,while Cyr61 reduced miR-195 expression and promoted the proliferation of JEG-3 cell respectively.Conclusions E2 and Cyr61 may play an important role in trophoblast cell proliferation by inhibiting the expression of miR-195,which provide a clue for exploring the pathogenesis of preeclampsia.
