摘要
以柠条锦鸡儿嫩枝为试材,观察了不同灭菌时间以及不同激素浓度对柠条外植体萌芽率、生根、愈伤组织诱导的影响,结果表明,柠条锦鸡儿茎段用0.1%升汞灭菌8 min时,污染率最低;以MS+0.2 mg/L 6-BA为培养基进行增殖培养时萌芽率最高,为80%;以1/2MS+0.5 mg/L IAA为生根培养基时生根率可达53%;以MS+0.5 mg/L 6-BA+2.0 mg/L 2,4-D为愈伤组织培养基时诱导率为63%,且愈伤组织出现早,生长良好。
In this paper,we took the Caragana korshinskii Kom.as experimental materials which were dealed under different sterilization time and different hormones,then we observed germination rate,rooting,callus tissue of the explants.The results showed that Caragana korshinskii Kom.had the lowest pollution when they were dealed by 0.1% HgCl2 in 8 min,they had the highest germination rate 80% when they were cultured by MS+ 0.2 mg/L 6-BA,and when they were dealed for rooting on 1/2MS+0.5 mg/L IAA,the rooting rate reached 53%,when the callus tissue medium was MS+0.5 mg/L 6-BA +2.0 mg/L 2,4-D,the callus tissue appeared earlier and growth in a good condition.
