大叶藓属(Rhodobryum)植物组织培养研究 被引量:3

摘要 研究以采自河北省境内的大叶藓(Rhodobryum roseum)为实验材料,探讨了不同培养基、不同培养条件对其配子体组织培养的影响,以期对大叶藓属植物组织培养研究和快速繁殖技术提供科学依据。结果表明:茎尖片段培养的原丝体和新芽生成比例明显高于叶片片段,采用茎段更适合于大叶藓的配子体组织培养;适度的Na-ClO溶液消毒外植体利于大叶藓配子体组织培养;6-BA抑制了大叶藓原丝体的生成,2,4-D则有明显的促进作用,6-BA促进了大叶藓愈伤组织和新芽的出现;BG11培养基更适于大叶藓原丝体的萌发和愈伤组织的生成,其他适于大叶藓组织培养的培养基依次为Knop、改良Knop、BBM、1/2MS和MS。 The influence of different medium and culture condition to the Rhodobryum roseum gametophyte tissue cultivation was studied in this article,which is expected to provide scientific basis to the tissue cultivation and the rapid reproduction techniques of Rhodobryum.The results indicate that the ratio of protonema germination and sprouting by the stem fragments is much more than that by the leaf fragments,therefore the stem fragments are more adequate for the gametophyte cultivation of Rhodobryum roseum.Sterilization by NaClO to the gametophyte fragments is good for the cultivation of Rhodobryum.6-BA inhibits the protonema development but promotes the callus and prolification,while 2,4-D promotes the protonema development.BG11 culture medium is the best for the gametophyte tissue cultivation,the other usable medium are Knop,modified Knop,BBM,1/2MS and MS.
