黑曲霉半固体发酵产单宁酶的工艺优化 被引量:1

Optimization of Tannase Production Technology from Aspergillus niger under Semisolid Fermentation

摘要 采用紫外分光光度法测定酶活,用单因素法对黑曲霉B0201半固体发酵产单宁酶的工艺条件进行优化。结果表明:半固体发酵法主要产胞内酶,3.3%五倍子、2.5%的泡沫、温度30℃、初始pH 5.0,接种量1×108个孢子,装液量60 mL,摇床转速160 r/min是产酶的最佳工艺条件,发酵72 h,单宁酶的活力最高,可达144.39 U/mL。该条件下半固体发酵所产胞内单宁酶酶活较高,工艺简单易控制。 The tannase activity was determined by ultraviolet spectrophotometry and the technological condition of tannase production from Aspergillus niger under semisolid fermentation was optimized.The results showed that the optimum technological conditions of tannase production from Aspergillus niger under semisolid fermentation were 3.3% gallnut,2.5% foam,initial pH5.0,inoculation amount of 1×10^8 spores,60mL medium,160r/min,30℃ and 72h.The tannase activity produced under the optimum technological condition can reach 144.39U/mL.
