摘要
目的:建立高效液相色谱法测定忍冬藤不同采收期中绿原酸和咖啡酸含量的方法。方法:色谱柱为UltimateTMXB-C18(4.6 mm×150 mm,5μm);流动相为0.3%-甲酸和乙腈(12∶88);流速为1.0 ml/min;检测波长为327 nm;柱温为25℃。结果:绿原酸在2.5~125.0μg/ml范围内呈良好线性关系;咖啡酸在1.2~60.0μg/ml范围内呈良好线性关系;加样回收率为96.0%~104.6%。结论:应用新建立的方法测定了不同采收期忍冬藤中绿原酸和咖啡酸2种活性成分的含量,该法简单、快速、准确,可用于忍冬藤和叶的质量控制。
Objective: To establish a method for the content determination of Chlorogenic acid and Caffeic acid in Caulis lonicerae during different collected periods by HPLC.Methods: The UltimateTM XB-C18 column(4.6 mm×150 mm,5 μm) was used.The mobile phases consisted of 0.3% formic acid and acetonitrile(12∶88).The flow rate was 1.0 ml/min and the detection wavelength was set at 327 nm,column temperature was 25℃.Results: The linear ranges of Chlorogenic acid was at 2.5-125.0 μg/ml and the linear ranges of that of Caffeic acid was at 1.2-60.0 μg/ml.The recovery rate was from 96.0% to 104.6%.Conclusion: The method is applied to the analysis of different collected periods Caulis lonicerae samples that came from different areas in China.The method is simple,rapid and accurate,and can be used for the evaluation of Caulis lonicerae.
