骨形态发生蛋白7在前列腺癌骨转移中的意义 被引量:2

Significance of bone morphogenetic protein 7 expression in osteoblastic metastasis of prostate carcinoma

摘要 目的 研究骨形态发生蛋白7(BMP7)与前列腺癌骨转移的关系.方法 采用逆转录-聚合酶链反应(RT-PCR)法检测人前列腺癌PC-3、乳腺癌细胞MDA-MB-231、肾癌细胞KETR-3、膀胱癌细胞T24、胃癌细胞MGC-803中BMP7 mRNA表达.免疫组织化学PV法检测45例有骨转移,42例无骨转移前列腺癌患者前列腺癌组织的BMP7蛋白表达.结果 RT-PCR结果显示,BMP7相对吸光度均值在前列腺癌PC-3细胞(9.59×10-1±1.67×10-2)和乳腺癌细胞(9.50×10-1±3.89×10-2)显著增高 与胃癌细胞MGC-803(2.93×10-1±1.33×10-2)、膀胱癌细胞T24(2.96×10-1±1.21×10-2)、肾癌细胞KETR-3(3.26×10-1±1.53×10-2)比较,差异有统计学意义(P<0.05).免疫组织化学结果显示,骨转移前列腺癌组BMP7表达吸光度均值(74.80±5.76)明显高于无骨转移前列腺癌组(65.94±1.76),两组差异有统计学意义(P<0.05).结论 BMP7在嗜骨性转移的前列腺癌细胞中,以及有转移的前列腺癌组织中的高表达,表明BMP7与前列腺癌骨转移有关. Objective To detect the expression of bone morphogenetic protein 7 (BMP7) in five carcinoma cells and prostate carcinoma tissues, and to explore the role of BMP7on osteoblastic metastasis of prostate carcinoma. Methods In vitro human prostate carcinoma cells PC-3, breast carcinoma cells MDA-MB-231, renal carcinoma cells KETR-3, bladder carcinoma cells T24 and gastric carcinoma cells MGC-803 were cultured, the total RNA were extracted from cells and the expression of BMP7 mRNA in cells were detected by reverse transcription-polymerase chain reaction (RT-PCR). Meanwhile, the expression of BMP7 in prostate carcinoma tissue samples were detected by Immunohistochemical PV method.Results RT-PCR results showed that the mRNA expression ratio of BMP7/GAPDH in human prostate carcinoma cells PC-3 was 9. 585 × 10-1 ± 1. 668 × 10-2, and breast carcinoma cells MDA-MB-231 was 9. 497 × 10 - 1 ± 3. 886 × 10 -2, there was no significant difference between them. The mRNA expression ratio of BMP7/GAPDH in human renal carcinoma cells KETR-3, bladder carcinoma cells T24 and gastric carcinoma cells MGC-803 were lower than that of human prostate carcinoma cells PC-3 and breast carcinoma cells MDA-MB-231. Immunohistochemical results showed that in prostate carcinoma without bone metastases the mean expression of BMP7 (65.94 ± 1.76) was significant lower in bone metastases(74. 80 ±5.76). Conclusion The BMP7 was overexpressed in the carcinoma cells and bone metastases of prostate carcinoma, which may play an important role in promoting osteoblastic metastasis of prostate carcinoma.
