猪苓及猪苓多糖对BBN诱导的膀胱癌大鼠外周血T淋巴细胞亚群表达的影响 被引量:20

Influence of Polyporus and Polyporus Polysaccharide on T-lymphocyte Subsets in Peripheral Blood of Bladder Cancer Rats

摘要 目的观察猪苓及猪苓多糖(PPS)对BBN诱导的膀胱癌大鼠外周血T淋巴细胞亚群分布的影响。方法药物干预膀胱癌大鼠第6周、第12周后眼眶后静脉丛采血,流式细胞术检测外周血T淋巴细胞亚群CD3+、CD4+和CD8+、CD28+、CD25+及TCRγδ+表达变化。结果模型组外周血CD3+、CD4+CD3+和CD8+CD3+、CD28+的T淋巴细胞的百分率(均值)均明显低于空白对照组;药物干预第6周后,PPS及不同剂量猪苓组CD8+CD3+和CD28+与模型组比较均显著升高(P<0.05);第12周后,PPS组CD4+CD3+的T淋巴细胞的百分率显著升高(P<0.05),不同剂量猪苓组CD28+表达均升高,其中中剂量组升高显著(P<0.05)。PPS组TCRγδ+的T淋巴细胞的百分率(均值)同模型组比较显著升高(P<0.05)。结论 PPS可通过上调膀胱癌大鼠外周血的CD8+CD3+、CD28+及TCRγδ+T淋巴细胞水平,增强膀胱癌大鼠对抗原的免疫应答水平,从而促进免疫功能的恢复。 Objectives To observe the influence of Polyporus and Polyporus Polysaccharide(PPS) on T-lymphocyte subsets in peripheral blood of BBN-induced bladder cancer rats.Methods Blood was collected from anaesthetized rats by orbital and cardiac puncture 6 and 12 weeks after medication.The flow cytometry was used to detect CD3^+,CD4^+,CD8^+ and TCRγδ^+T cells in peripheral blood of different groups.Results The percentages of CD3^+,CD4^+ CD3^+,CD8^+ CD3^+ and CD28^+ in peripheral blood of BBN model group was significantly lower than those in the blank control group.After treatment with different does of Polyporus and PPS for six weeks,the percentages of CD8^+CD3^+and CD28^+ were markedly increased(P〈0.05).After treatment with PPS for twelve weeks,the percentage of CD4^+CD3^+,and TCRγδ^+were significantly increased(P〈0.05).CD28^+ percentage was increased in Polyporus groups,particularly in middle-dosage Polyporus group(P〈0.05).Conclusion PPS can promote the recovery of immune function probably through up-regulating the expression of CD8^+CD3^+and CD28^+ and TCRγδ^+and enhancing the ability of immune response to antigens.
