摘要
目的通过建立KM种小鼠实热壅滞的粪性腹膜炎(ABP)模型,观察大承气汤对实热壅滞的粪性腹膜炎小鼠内毒素及血清丙二醛(MDA)和超氧化物歧化酶(S0D)含量的影响,探索大承气汤治疗急腹症类疾病的机理。方法 KM小鼠64只,随机分为对照组、实热壅滞的粪性腹膜炎模型组、大承气汤治疗组和氨苄西林治疗组,应用专属试剂盒分别检测各组外周静脉血及回肠内容物的内毒素含量,测定血清MDA、SOD含量。结果造模各组小鼠出现口唇发绀、眼角分泌物浓稠、竖毛、少动、蜷曲蹲伏、不进食、大便极干、腹部鼓胀等症状。解剖腹腔可见腹腔内有少量浑浊液体,肠管充血,多数肠管膨胀,部分肠管痉挛。大承气汤组与模型组相比,外周血及回肠内容物中的内毒素含量、血清MDA含量均明显降低(P<0.05),SOD含量显著升高(P<0.01),但氨苄西林组与模型组相比上述指标差异不明显(P>0.05)。结论大承气汤能抑制实热壅滞的粪性腹膜炎小鼠内毒素位移和氧化-抗氧化失衡。
Objective To investigate the effect of Da Chengqi Decoction(DCD) on the endotoxin translocation and the content of malondiadehyde(DMA) and superoxide dismutase(SOD) in serum of mice with acute bacterial peritonitis(ABP).Methods Sixty-four KM mice were divided into four groups randomly: control group,ABP group,DCD group and ampicillin group.We detected the content of endotoxin in the peripheral blood and ileal contents.The serum contents of MDA and SOD in each group were also examined.Results In ABP group,mice presented the manifestations of oral cyanosis,canthal sticky excretion,hair erecting,less movement,crimping and squatting,apastia,extremely dry stool,abdominal tympany.The results of anatomical examination showed that a small amount of turbid liquid was seen in the abdominal cavity,the intestines were congested,most of the intestinal canal was dilated,some of the intestinal canal was in spasm.Compared with those in ABP group,serum MDA and endotoxin in the peripheral blood and ileal contents were significantly decreased in DCD group(P〈0.05),and serum SOD was increased(P〈0.01).No significant difference was shown between ampicillin group and ABP group(P〈0.05).In ampicillin group,serum MDA was negatively correlated with serum SOD(P〈0.05).Conclusion DCD can inhibit endotoxin translocation and regulate the oxidant/antioxidant imbalance in ABP mice.
