摘要
目的研究尪痹片对热杀死结核分枝杆菌(Mtb)诱导的SD大鼠佐剂性关节炎踝关节组织病理损伤的抑制作用。方法除正常组外,其余各组大鼠用Mtb与矿物油混悬液一次性经尾根部皮下注射,建立Mtb诱导的佐剂性关节炎大鼠模型。将模型大鼠随机分为模型对照组(生理盐水2 mL)、尪痹片高、低剂量组(3.2,1.6 g/kg)、正清风痛宁组(4.25 mg/kg),正常对照组给予生理盐水2 mL。每天灌胃给药1次,连续4周。给药结束后,取大鼠踝关节进行病理检查,观察大鼠踝关节滑膜、软骨、关节腔、周围软组织炎症反应等组织病理改变情况,进行综合评分。结果与模型对照组比较,尪痹片能够显著抑制炎症细胞的浸润和滑膜增生,减轻软骨损伤程度(P<0.05)。结论尪痹片对实验性类风湿关节炎的滑膜及关节软骨具有较好的保护作用。
Objective To investigate the inhibitory action of Wangbi Pellet(WP) on ankle damages in SD rats with adjuvant arthritis(AA) induced by heat killed mycobacterium tuberculosis H37RA(Mtb).Methods Except for the normal control group,all rats were injected with one-dose of the mixture of Mtb(1mg per rat) with mineral oil by subcutaneous injection through the base of the tail to induce the model of AA.SD rats with AA were randomly divided into four groups: model group(gastric gavage of normal saline 2 mL per rat),high-and low-dose WP groups(gastric gavage of WP 3.2 g/kg,1.6 g/kg,respectively),positive control group(gastric gavage of Zhengqing Fengtongning 4.25 mg/kg) and normal control group(gastric gavage of saline 2 ml per rat).The rats in each group were respectively treated with corresponding medicine once a day for 4 weeks.The ankle tissue was examined under microscope to evaluate the pathological changes of synovium,cartilage and articular cavity as well as soft tissue inflammation.Results WP remarkably inhibited the hyperplasia of synovial membrane and the infiltration of inflammatory cells,and reduced cartilage injury as compared with the model group(P〈0.05).Conclusion WP has certain protective effect on ankle synovium and cartilage of rats with experimental arthritis.
