狭基线纹香茶菜保肝、利胆的实验研究

Studies on Liver-protective and Choleretic Effect of Isodon lophanthoides var.gerardianus

目的研究狭基线纹香茶菜对酒精性肝损伤、胆汁瘀积型肝损伤等模型动物的保护作用。方法建立小鼠急性酒精中毒致肝损伤模型,检测药物对小鼠血清丙氨酸氨基转氨酶(ALT)、天冬氨酸氨基转移酶(AST)活性,肝组织中丙二醛(MDA)含量及肝脏指数的影响;建立大鼠慢性酒精性脂肪肝模型,观察药物对大鼠死亡率,肝脏病理损伤程度及肝、胸腺指数的影响;观察药物对大鼠胆汁分泌量的影响;建立大鼠胆汁瘀积型肝损伤模型,观察药物对其死亡率的影响。结果狭基线纹香茶菜提取物能显著降低急性酒精中毒小鼠血清ALT、AST含量和慢性酒精性脂肪肝大鼠的死亡率,减轻肝脏组织的病理损伤程度;能促进正常大鼠的胆汁分泌,但对胆汁瘀积型肝损伤的作用不明显。结论狭基线纹香茶菜能够较好地减轻酒精性肝损伤,且具有一定的利胆作用。

Objective To study the protective effect of Isodon lophanthoides var.gerardianus on alcohol-induced and cholestasis-induced liver injury.Methods Acute alcohol liver injury model was made in mice by an oral administration of alcohol（56°,14 mL·kg-1）,and then we observed the effects of Isodon lophanthoides var.gerardianus on serum glutamic pyruvic transaminase（ALT） and glutamic oxaloacetic transaminase（AST） activities,liver malondia1dehyde（MDA） and organ indexes of liver.Chronic alcoholic-fatty liver model was induced by 50 % alcohol and high-fat feeding in rats,and the effects of Isodon lophanthoides var.gerardianus were evaluated by mortality and hepatic histological changes.Drug-stimulated bile secretion was determined in anesthetized rats.Acute cholestatic injury model was induced by bile duct ligation,and the effects of Isodon lophanthoides on this model were evaluated by mortality.Results The aqueous extract of Isodon lophanthoides var.gerardianus could significantly reduce the ALT and AST levels in alcohol-induced acute liver injury mice,decrease the mortality of chronic alcoholic fatty liver rats and alleviate liver injury.The extract could also stimulate the bile secretion in rats but had no effect on cholestasis-induced liver injury.Conclusion The aqueous extract of Isodon lophanthoides could attenuate alcohol-induced liver injury in mice or rats and improve the bile secretion in rats.
