摘要
探讨了头孢唑啉钠、头孢曲松钠、头孢霉素3种头孢类抗生素对农杆菌EHA105的抑制作用,以及对中黑防杨叶片生长和分化的影响。结果表明:头孢霉素和头孢曲松钠的抑菌效果优于头孢唑啉钠,但二者对叶片分化的抑制性太强,而750 mg/L的头孢唑啉钠对叶片分化影响很弱,仅比对照低3%,同时抑菌效果好。因此确定了由农杆菌介导的中黑防2号杨遗传转化时的抗生素为头孢唑啉钠其使用浓度为750 mg/L。
This paper discusses the cefazolin sodium,ceftriaxone sodium,and cefotaxime,three kinds of cephalosporin antibiotics on the inhibition of agrobacterium EHA105,and effect on the growth and differentiation of Populus deltiodes × cathayana leaves.The results showed that: cefotaxime and ceftriaxone sodium are superior to the inhibitory effect of cefazolin sodium,but the leaves differentiation inhibition caused by the two pairs of cephalosporin antibiotics is too strong,while 750 mg/L of cefazolin sodium has very weak influence on the differentiation of leaves,only 3% lower than that of comparison,and inhibitory effect is satisfactory.Thus it is determined that the genetic transformation antibiotics of agrobacterium-mediated Populus deltiodes×cathayana is cefazolin sodium,whose concentration for use is 750 mg/L.
