摘要
目的观察槐定碱(sophoridine,SRI)致痫大鼠行为学、大鼠海马CA3区神经元组织形态学和超微结构,探讨槐定碱毒理作用。方法将30只成年SD大鼠(雌雄不拘)随机分为生理盐水对照组(NS)、槐定碱组(SRI)、苯巴比妥钠(槐定碱组(PBS+SRI),每组各10只。腹腔注射大剂量槐定碱(47.83 mg/kg)后,观察大鼠行为学变化以及应用电镜技术观察各组大鼠致痫后海马CA3区神经元组织病理学和形态学的改变。结果腹腔注射大剂量槐定碱后,大鼠出现了痫性行为发作;电镜下可见槐定碱致痫大鼠海马CA3区神经元核膜双层结构破坏,神经细胞核严重固缩,染色质颗粒状聚集,线粒体肿胀,嵴断裂或消失,毛细血管内皮细胞核固缩。经苯巴比妥钠干预后,槐定碱致痫大鼠海马CA3区神经细胞的损伤程度减轻。结论大剂量槐定碱可导致大鼠在体海马神经细胞损伤,从而诱发癫痫。
Objective To observe the ultrastructure and morphological deformation of hippocampus in SRI-induced epileptic rat.Methods Sprague Dawley rats were randomly divided into saline control group(n=10),SRI group(n=10) and sodium phenobarbital(PBS)+ SRI group(n=10).SRI was injected into the abdominal cavity of rats to observe the changes of rat behavior,then ultrastructure and morphological deformation of neurons in CA3 of hippocampus in saline control group,SRI group and sodium phenobarbital(PBS)+ SRI group rats were observed through HE staining and electronic microscope.Results The kindling model of rats was induced by injection of SRI intraperitoneally.The behavioral seizure was observed,sings of mitochondrial damage,organelle burst and degeneration of synapses were seen after SRI administration;Neurons of PBS+ SRI group were less damaged as compared with those of SRI group and most neural structure was not damaged.Conclusion Sophoridine can cause hippocampal neurondamage and induce seizure in vivo.
