The expression levels of TNF α mRNA in synoviocytes from adjuvant arthritis rats (AA) were measured by semiquantitative RT PCR.The results showed that the production of TNF α mRNA by synoviocytes from AA rats were greatly enhanced as compared with that of normal controls ( P <0.01).After treatment with Tongbiling (TBL),the expression levels of TNF α mRNA in synoviocytes were significantly decreased ( P <0.01).The effects could be observed in methotrexate treated grop ( P <0.05),but less than in TBL treated grop.Whereas indomethacin further enhanced the expression levels of TNF α mRNA in synoviocytes ( P <0.05).Our data suggest that abnormal expression of TNF α mRNA in synoviocytes plays an important role in the pathogenesis of AA rats.The mechanisms of the therapeutic effect of TBL on AA might be due to inhibit the expression levels of TNF α mRNA in synoviocytes,and then to decrease the production of TNF α.
