摘要
[目的]研究不同浓度NAA和6-BA组合的MS培养基对月见草种子发芽和愈伤组织诱导的影响。[方法]通过组织培养,研究了NAA和6-BA16种组合的MS培养基中的月见草种子发芽率和愈伤组织诱导率。[结果]在NAA和6-BA的16种组合中,MS+10mg/L6-BA培养基中的种子发芽率最高(90%),其次是MS+10mg/L6-BA+2mg/LNAA(87%),再次是MS+2mg/L6-BA(83%)和MS+10mg/L6-BA+5mg/LNAA(83%);MS+2mg/L6-BA+5mg/LNAA培养基中的愈伤组织诱导率最高(88.89%),其次是MS+5mg/L6-BA+10mg/LNAA(87.50%)、MS+2mg/L6-BA+2mg/LNAA(85.71%)和MS+2mg/L6-BA+10mg/LNAA(83.33%);MS+10mg/LNAA和MS+5mg/LNAA培养基中有不定芽和不定根产生。[结论]该研究为提高月见草种子发芽率和愈伤组织诱导率提供了技术指导。
[Objective]The aim was to study effects of MS mediums with different combinations of NAA and 6-BA on seed germination and callus induction of evening primrose.[Method]Germination rate and callus induction rate of evening primrose seed in MS mediums with different combinations of NAA and 6-BA were studied by tissue culture.[Result]In 16 combinations of NAA and 6-BA,MS+10 mg/L 6-BA medium had the highest germination rate of seed (90%); next came MS+10 mg/L 6-BA+2 mg/L NAA medium (87%),MS+2 mg/L 6-BA medium (83%) and MS+10 mg/L 6-BA+5 mg/L NAA(83%).MS+2 mg/L 6-BA+5 mg/L NAA medium had the highest callus induction rate (88.89%); next came MS+5 mg/L 6-BA+10 mg/L NAA medium (87.50%),MS+2 mg/L 6-BA+2 mg/L NAA medium (85.71%) and MS+2 mg/L 6-BA+10 mg/L NAA medium (83.33%).There were adventitious buds and adventitious roots in MS+10 mg/L NAA and MS+5 mg/L NAA mediums.[Conclusion]The research provides the technical guidance for improving seed germination and callus induction of evening primrose.
