急性心肌梗死患者单核细胞PPARδ、MCP-1和MMP-9表达及其与冠状动脉病变关系 被引量:12

Expressions of peroxisome proliferator-activated receptor δ,monocyte chemoattractant protein-1 and matrix metalloproteinase-9 in monocyte and their correlations with coronary artery lesions in acute myocardial infarction

摘要 目的:探讨急性心肌梗死患者单核细胞中过氧化物酶体增殖激活受体δ(peroxisome proliferator-activatedreceptorsδ,PPARδ)、单核细胞趋化蛋白-1(monocyte chemoattractant protein-1,MCP-1)和基质金属蛋白酶-9(matrixmetalloproteinase-9,MMP-9)的表达及与冠状动脉病变的关系。方法:46例急诊行冠状动脉介入治疗的急性心肌梗死患者和46例冠状动脉造影排除冠心病者(对照组),分离其周围血单核细胞,应用蛋白免疫印迹法检测PPARδ、MCP-1和MMP-9的表达;分析其与冠状动脉病变程度的相关性。结果:急性心肌梗死患者血单核细胞中PPARδ表达水平低于对照组(P<0.01),MCP-1及MMP-9表达高于对照组(P<0.01);PPARδ表达水平与冠状动脉病变程度呈负相关,MCP-1、MMP-9与冠状动脉病变程度呈正相关,PPARδ表达水平与MCP-1、MMP-9呈负相关。结论:PPARδ可通过抑制炎症介质MCP-1及MMP-9的表达,发挥抗动脉粥样硬化及稳定粥样斑块的作用。 Objective To investigate the expressions of peroxisome proliferator-activated receptor δ,monocyte chemoattractant protein-1 and matrix metalloproteinase-9 in peripheral blood monocytes and their correlations with their coronary artery lesions in acute myocardial infarction.Methods Peripheral blood monocytes were collected from 46 patients with acute myocardial infarction and 46 subjects without acute myocardial infarction detected with angiography.The expressions of peroxisome proliferator-activated receptor δ,monocyte chemoattractant protein-1 and matrix metalloproteinase-9 were detected with Western blotting,and their correlations with the severity of the coronary atherosclerotic lesions were analyzed.Results The peroxisome proliferator-activated receptor δ level was lower,and monocyte chemoattractant protein-1 and matrix metalloproteinase-9 levels were significantly higher in patients with acute myocardial infarction than those in control group(all P0.01).The peroxisome proliferator-activated receptor δ level was negatively while the monocyte chemoattractant protein-1 and matrix metalloproteinase-9 levels were positively correlated to the severity of the coronary artery lesions.The peroxisome proliferatoractivated receptor δ level was negatively correlated to the monocyte chemoattractant protein-1 and matrix metalloproteinase-9 levels.Conclusion The peroxisome proliferator-activated receptor δ might play a role in the prevention of atherosclerosis and in the stabilization of the vulnerable plaque by inhibiting the expressions of inflammatory mediators monocyte chemoattractant protein-1 and matrix metalloproteinase-9.
